My wife always gets Caesar Salad with chicken no matter where she goes. Our server Haley did a great job and we got our drink order right away. We got a real nice inside seat right next to the river and got waited on right away. I used to go there years ago with my son and it was great. thought we would give them another try since they are on the water. Our favorite Italian restaurant was closed yesterday so I. The "music" was blasting and nobody even wanted to turn it down. My wife and I stopped there several times in the past and we walked in the front door and turned around and walked out. We really had a disappointing dinner and couldn’t wait to leave which is usually the opposite feeling one has when sitting on the deck with a beautiful waterfront view. The clam basket was mostly fries with little clams. The food came out promptly and was just “fine”. The ice was melted and it looked much more like a Shirley Temple than what was ordered. My husband ordered an Old Fashioned to drink and after a significant wait, someone (not our server) brought it to us, complaining that “she didn’t know where our server had gotten off to” and left the drink. Additionally there was music already being piped onto the deck so it was very overwhelming to have both of these hitting you while trying to eat and enjoy the scenery. We were outside on the far end, furthest away from the band and yet it was still extremely loud. Friday night and shortly after being seated the live music began. On the bright side, everyone was very nice and the view was beautiful. We were excited to visit this restaurant as we love it’s sister establishment across the water.

The report cites an anonymous source, however, who said that Evernote is in a “death spiral” due to flat user growth and a lack of enterprise adoption:Ī person who tipped TechCrunch off to the executive departures gave a slightly more blunt and uncharitable spin on the state of affairs. While Evernote confirmed the departure of each executive to TechCrunch, the company did not offer any reasoning. Ranjit Probhu, who joined Evernote in May of this year as a SVP of engineering, will partner with Andrew Malcom, SVP of product, growth, and marketing, to work on “how tech and product will fit together.” Susan Stick, who was hired in June as Evernote’s general counsel, is expanding to cover “people operations.” Francie Strong, VP of communications, is also expanding to be SVP of brand and communications. According to a new report from TechCrunch, Evernote has lost its CTO, CFO, CPO, and HR head in the last month…Īmid the departure of CTO Anirban Kundu, CFO Vincent Toolan, CPO Erik Wrobel and head of HR Michelle Wagner, Evernote has moved several recent hires to expanded roles within the company. Evernote has slashed the price of its Premium membership as a new report indicates the company is struggling financially and facing leadership departures. The research described herein conformed to the Helsinki Declaration. Additionally, these libraries may contain fragments that share sequence similarity to recombined TCR sequences (e.g., the red transcript), potentially leading to false-positives The resulting sequencing library will contain fragments that, by chance, contain the CDR3 encoding sequence. c RNA-seq employs shotgun sequencing, generating fragments from all transcripts present in the sample, which then have sequencing adapters ligated ( black). b TCR-seq involves selective amplification of the CDR3 region of TCR transcripts (displayed as a color gradient) by reverse transcription polymerase chain reaction (PCR) shown using a conserved C-gene primer ( purple with black sequencing adapter tails) for the initial reverse transcription step and resulting, after PCR (not shown), in an enriched set of recombined TCR sequences. a A pool of all mRNA in a sample is depicted, which contains irrelevant transcripts ( blue, brown, and red) as well as recombined TCR transcripts ( multi-colored). Each color represents a unique gene sequence. Horizontal lines represent mRNA transcripts with gray poly-A tails. Schematic representation of T cell receptor sequencing ( TCR-seq) versus RNA sequencing. This necessitates an analytical approach that is both fast and accurate for TCR extraction from tissue-derived RNA-seq datasets. For a pure lymphocyte population, only one in approximately 2000 transcripts are TCR transcripts (see “ Methods”) and in tissues T cells represent a minor cell type, further decreasing TCR transcript representation. Compared to TCR-seq, the main challenge in CDR3 extraction from tumor RNA-seq data is the disproportionally large number of non-TCR transcripts (Fig. Here, we describe an optimized approach for TCR CDR3 extraction from RNA-seq datasets from solid tumors, for the purpose of characterizing T cell populations present in the tumor environment. Next-generation sequencing technology has made whole-genome and transcriptome sequencing routine, and provided opportunities for the extraction of immunological data, such as human leukocyte antigen (HLA) type, using specialized software tools. However, because they rely on targeted amplicon sequencing, they do not evaluate TCR variation in the context of the overall genetic diversity of the specimen from which the data are derived. Ĭonventional TCR-seq methods provide a detailed view of TCR diversity. TCR-seq applied to tissue specimens can provide insight into tumor-infiltrating lymphocytes, T cells associated with autoimmune pathology and infection, and the properties of normal primary and secondary lymphatic tissues. Typically, CDR3 sequence information is acquired by performing TCR-sequencing (TCR-seq) experiments on peripheral T cells isolated from blood amplifying the CDR3 region with a conserved C gene primer followed by 5′ rapid amplification of cDNA ends, or with a multiplexed set of V and J gene primers. CDR3 is the TCR motif that directly binds MHC-presented peptide epitopes and this binding interaction is the main factor conferring T cell antigen specificity. T cells recognize peptide epitopes presented on the surface of cells on major histocompatibility complex (MHC) molecules. Primary sequence analysis of the highly variable complementarity determining region 3 (CDR3) of rearranged T cell receptor (TCR) genes provides insight into the adaptive immune response.

One was in Puyallup, Washington, and another in Arizona. Of the fifteen centers, twelve were in California. The complexes were large enough to house thousands of people and flexible enough to change functions quickly. These temporary detention centers were mostly repurposed facilities like racetracks and fairgrounds. Officials of the Wartime Civil Control Administration, a branch of the Western Defense Command, ordered people to report to fifteen “assembly centers” along the West Coast. Nevertheless, by the end of March, DeWitt had begun to forcibly remove about 120,000 Japanese Americans from their homes and businesses. Lack of contacts outside the exclusion zone, frozen bank accounts, and hostility from inland communities contributed to west coast Japanese Americans' reluctance-and in many cases, inability-to move.īetween 19, no person of Japanese ancestry was ever charged with spying. An initial voluntary evacuation had relatively little effect. With its new powers, the US military ordered all people of Japanese ancestry, citizens or otherwise, excluded from Military Area 1. The remainder of those western states comprised Area 2. Both were massive: Area 1 included the entire West Coast-Washington, Oregon, and California-from the Pacific Ocean inland to approximately the eastern side of the Cascades mountain range, as well as the southern half of Arizona. DeWitt, head of the Western Defense Command, issued Public Proclamation No. In practice, the order signaled a large-scale, interstate program that relocated and imprisoned Japanese Americans. The order is abstract: any location, any size, any restrictions. Within that area, the normal rights of American citizens would no longer apply.Įxecutive Order 9066 does not specifically mention people of Japanese or any other particular background. Within that area, the military could force you to stay or force you to leave. In other words, the president gave his military commanders the power to draw a line around any part of the country. In the order, the president authorized and directed the Secretary of War “to prescribe military areas in such places and of such extent as he may determine, from which any or all persons may be excluded, and with respect to which, the right of any person to enter, remain in, or leave shall be subject to whatever restrictions the Secretary of War may impose in his discretion.” The next day, the United States declared war against Japan. About two months earlier, the Imperial Japanese Navy had attacked Pearl Harbor. On February 19, 1942, President Franklin D. You just have to stay on until the end of the line. But first, there's another important stop. If you visit the Nikkei Center, you'll find a powerful permanent exhibit that begins to tell the story of how and why this part of Portland's identity changed. In Portland today, Japantown is no longer Japantown, except in the memories of some citizens and in the records preserved in the archives of the Oregon Nikkei Legacy Center, just steps from the tracks. Some of Old Town's blocks were once Portland's Japantown, though passengers on this MAX train won't see much sign of that history as they ride through. The next few stops are in Old Town, a National Historic Landmark district that also contains blocks Portlanders have known for decades as Chinatown. The train moves north through downtown Portland, crossing West Burnside Street. Its destination is no longer Portland City Center, but the Expo Center-across the Willamette River and along Interstate 5 through North Portland to the far northern edge of the city, nearly all the way to the Columbia River. Just like that, the Orange Line train has become a Yellow Line train. As the train continues north along the transit mall through downtown, the sign in the window of the train changes, as well as the color of the square displayed on the front. From May to September 1942, this site housed the Portland Assembly Center, where Japanese Americans from Oregon and Washington were held while the Minidoka War Relocation Center in Idaho was completed.Īn Orange Line MAX light-rail train leaves the station at Portland State University.
